A total of 48 healthy volunteers were randomly assigned to receive either 100 mg sildenafil or a placebo tablet (1mg/kg of body weight) for 4 days (control) or for a period of 4 days (the first 4 days of the study) with a daily dose of 50 mg sildenafil. The study was conducted between days 4 and 14 of the study.
At the end of the study, the erectile function of the men was assessed using the International Index of Erectile Function (IIEF) and the Prostate Index, as well as the International Index ofitement for Erectile Function (IIEF-EF). Results
The sildenafil group had significantly higher levels of cGMP in the corpus cavernosum of the men than in the placebo group (P<0.001). The plasma concentration of cGMP was significantly greater in the sildenafil group than in the placebo group (P<0.05). The plasma levels of sildenafil and the plasma concentrations of sildenafil and placebo were significantly higher in the sildenafil group than in the placebo group (P<0.001 and P<0.001, respectively).
Further, the plasma concentrations of sildenafil and the plasma concentrations of sildenafil and placebo were significantly lower in the sildenafil group than in the placebo group (P<0.001 and P<0.001, respectively).
A significant decrease in the plasma levels of sildenafil and the plasma concentrations of sildenafil and placebo was observed in the sildenafil group compared with the placebo group (P<0.001 and P<0.001, respectively).
